LGD-4033

A randomized controlled trial found LGD-4033 to be well-tolerated in 76 healthy men after receiving dosages of 0.1, 0.3, or 1 mg per day for 21 days. The Journals of Gerontology concluded that LGD-4033 “was safe” despite observing reductions in high-density lipoprotein (HDL) and testosterone. They reported no changes in AST or ALT (Basaria et al., 2013).

Research has shown that drinking carbonated water can increase HDL and decrease LDL levels, with carbon dioxide acting as a vasodilator. Therefore, consuming carbonated water is recommended for individuals using LGD-4033 or other SARMs.

RAD-140

Clinical Breast Cancer researchers found RAD-140 to have an “acceptable safety profile” following doses of 50 mg, 100 mg, and 150 mg per day. However, raised liver enzymes, vomiting, dehydration, and decreased appetite were reported as some of the side effects (LoRusso et al., 2022).

Ostarine

YK-11

A Chemico-Biological Interactions study in rats found that 0.35 g/kg of YK-11 altered hippocampal neurochemistry, resulting in memory impairment (Dahleh et al., 2024). Thus, YK-11 exhibits neurotoxic effects, causing damage to the brain. The researchers also observed increases in the cytokines IL-1β and IL-6 following YK-11 administration. These are small proteins that influence immunity, indicating heightened inflammation and oxidative stress.

For researchers seeking to reduce neurotoxicity during YK-11 cycles, they may supplement with benfotiamine. Benfotiamine is a highly bioavailable form of vitamin B1 that:

  • Decreases oxidative stress
  • Improves nerve conduction
  • Inhibits neurodegeneration

MK-677

Elsevier reported that 25 mg/day of MK-677, administered over two 14-day periods, did not cause any notable side effects (Murphy et al., 1998). It was described as “generally well-tolerated.”

Cardarine

When administered in high dosages and for extended durations, this peroxisome proliferator-activated receptor delta (PPARδ) agonist has been shown to cause cancer in mice. In other studies, doses of 10 mg/kg and 30 mg/kg of Cardarine inhibited tumorigenicity in female mice with nasopharyngeal carcinoma, a type of throat cancer.

Frontiers in Pharmacology states that Cardarine achieved this effect by suppressing cell division at the G2/M phase and promoting apoptosis, otherwise known as cell death (Ji et al., 2018). Currently, there is a lack of data regarding the carcinogenic effects of Cardarine in humans.

Cardarine users may support their immune health by supplementing with vitamin D3 (cholecalciferol). Vitamin D3 has been shown to promote anticarcinogenesis by enhancing the immune system’s production of killer cells, also known as T cells and NK cells. Administering 4,000 IU (100 mcg) of vitamin D3 daily has proven effective in reducing the incidence of illness among our patients.

S4

S4 can cause transient ocular changes in some patients, resulting in a yellow tint to their vision. However, permanent vision alterations following supplementation discontinuation are uncommon.

Researchers utilizing S4 may counteract vision deterioration by enhancing eye health with the following supplements:

  • Vitamin A: 3,000 IU (900 mcg) per day
  • Lutein: 10 mg per day
  • Zeaxanthin: 2 mg per day

S23

In preclinical research published in Endocrinology, S23 temporarily impaired spermatogenesis (sperm production) in rats. This effect occurred due to S23 decreasing luteinizing hormone (LH) and follicle-stimulating hormone (FSH) (Jones et al., 2009). This effect was shown to be reversible following S23 treatment, which is promising for safety.

He later took a plea deal, was suspended for one year, and received a fine of $1.1 million.

Joakim Noah was banned by the National Basketball Association (NBA) for 20 games after he was found taking LGD-4033. Noah said he regretted taking the SARM and that it was an ethical mistake on his part.
